PRODUCTION OF DIOXIN BIOSENSOR CELL LINE BY USING H4IIE CELL LINE

Abstract
The dioxin compounds impact on the cell through a specific protein - aryl hydrocarbon receptor (AhR) in cytoplasm and DRE domain of DNA, they regulate many different genes. Upon this mechanism we have done transferred plasmid pGudluc1.1 into H4IIe cell line expressed AhR protein in order to generate dioxin biosensor cell line. Plasmid pGudluc1.1 contains DRE domain that is located in the region of luciferase synthesis. We detect dioxin compounds by luciferase assay reaction right after biosensor cell line exposed to samples. The results showed that, a dioxin biosensor cell line was generated from H4IIe cell line and that can be applied in detection of dioxin compounds.
