PROTECTIVE EFFECT OF Pithecellobium dulce (roxb.) Benth. ROOT EXTRACT AGAINST TUNICAMYCIN-INDUCED CELL DEATH IN MIN6 PANCREATIC β-CELLS

Abstract
Objectives: Protective effect of Pithecellobium dulce (Roxb.) Benth. root extract against cell death was conducted in MIN6 pancreatic β-cells. Methods: The MIN6 cell line was cultured with 2.5 - 5 μg/mL tunicamycin (tm) added following 2, 24 and 48 hours of exposure to induce cell death. The protective effect of Pithecellobium dulce root extract against cell death was measured through various concentrations (50 to 500 μg/mL). Results: the optimal concentration of tunicamycin to cause cell death was 5 μg/mL in 24-48 hours of incubation. All tested concentrations of Pithecellobium dulce root extract had no toxicity towards the MIN6 cells during 48 hours of exposure. Concentrations 50 μg/mL and 100 μg/mL of the root extract possessed the protective effect against cell death in MIN6 cells with 24 and 48 hours of exposure, respectively. Conclusion: It is proved that Pithecellobium dulce root has potential in diabetes treatment by against cell death in pancreatic β-cells through tunicamycin.
