ACTIVITY XANTHINE OXIDASE INHIBITION OF ETHANOLIC EXTRACT FROM Achyranthes aspera L.

Abstract

Xanthine oxidase is an enzyme responsible for catalyzing the oxidation of hypoxanthine to xanthine and of xanthine to formation of uric acid. Objective: This study aimed to identify the xanthine oxidase inhibitors and decrease of serum uric acid levels from extract ethanol Achyranthes aspera in hyperuricaemic. Achyranthes aspera extract were prepared by maceration method in ethanol 70%. The xanthine oxidase inhibitory activity of extract ethanol Achyranthes aspera was determined by the level of uric acid formed. Methods: The concentration of uric acid was determined by Spectrophotometer method. Results: methanol extract of Achyranthes aspera have shown promising inhibited xanthine oxidase activity with IC₅₀ = 17.5 µg/mL, significant decreased in the serum urate level (186; 129.4 and 128.7 µmol/L), and reduced of xanthine oxidase activities in the mouse liver (2.62; 1.79 and 1.87 mM acid uric /min/g protein) at dosage 1.25 and 2.5/kg wb, respectively. Conclusion: Achyranthes aspera extract inhibits xanthine oxidase activity and decreases the plasma uric acid concentration.
