DEPLOYING THE MODEL OF OVARIAN FAILURE BY TRIPTERYGIUMGLYCOSIDE (TG) SUBCUTANEOUS INJECTION

Abstract

Objective: Ovarian failure model was induced in female mice with subcutaneous
tripterygium glycoside (TG). Methods: 6-week-old female Swiss mice were injected
subcutaneously TG 50mg/kg/day (0.1 ml/10g) for 35 consecutive days to cause ovarian failure. The control group was injected with distilled water. Evaluation of mice body weight, oestrus cycle, FSH and estrogen serum concentration, ovarian and uterus index, HE stained histopathology of uterine and ovarian, immunohistochemical staining of ovaries for Notch1 protein expression. Results: Compared with the control group, mice in TG injection group had increased rate of abnormal estrous cycle, decreased frequency of oestrus cycle (p < 0.01); increased serum FSH concentration, decreased serum E2 concentration (p < 0.01); decreased uterine and ovarian index (p < 0.01); HE stained histopathology of ovarian with decreased follicle growth and luteum, uterine with thinner endometrium and decreased glands; immunohistochemical staining of ovaries has a marked decrease in expression of the protein Notch1 (which plays an important role in endothelial development and balance by regulating cell proliferation, diferentiation and death). Conclusion: Successfully deployed a model of ovarian failure in female Swiss mice by injecting TG (Tripterygiumglycoside) 50mg/kg/day (0.1 ml/10g) for 35 consecutive days.
